从应用方向来看,依然涉及到了多个应用,而小分子应用文献数量超过了1/3,是前五个月文章中最多的应用,这个与一年前的统计比较类似(畅享小分子互作分析!)。足以说明Octet®不仅可以灵敏地测试小分子,而且简单易用,可以在更短时间内获得准确的动力学数据。今天陈老师就带大家继续畅游最近的优秀国内小分子文章。

微生物天然产物PROTAC分子[1]


该研究发现了第一个来源于微生物天然产物的PROTAC分子(蛋白降解靶向联合体)APL-16-5,同时结合 E3 泛素连接酶 TRIM25 和流感病毒 PA 亚基,基于 PROTAC 机制,诱导TRIM25依赖的PA泛素化及其降解,阻断病毒RNA复制,具有良好的体内抗甲型流感病毒活性。表明微生物代谢产物可成为发现天然PROTAC分子的重要来源。Octet®用来检测APL-16-5与TRIM25和PA的结合。

进入临床的降脂药[2]


小分子 DC371739在多种动物模型中能够同时显著降低血浆中总胆固醇、低密度脂蛋白胆固醇和甘油三酯水平,并且在1期临床试验(NCT04927221)中显示出初步阳性效果和良好的耐受性。从机制上来说,DC371739与其他已知的降血脂药物不同,它是通过直接与 HNF-1α 结合来阻止 PCSK9 和 ANGPTL3 的转录,从而发挥降血脂作用。Octet®检测HNF-1α与药物的结合。

高亲和力小分子抑制剂[3]


BET(Bromodomain and Extra-Terminal Domain)家族成员包括BRD2、BRD3、BRD4和BRDT,每个家族成员都具有两个N端串联的溴结构域(BD1和BD2)。BET蛋白作为转录调控的阅读器,与癌症、炎症等多种疾病的发生发展密切相关。第一代BET抑制剂(Pan BET)对BD1和BD2溴结构域不具备选择性,导致在临床试验中出现血小板减少、腹泻、恶心呕吐等毒副作用。本研究采用基于结构的药物设计策略,结合结构生物学研究开展了深入的构效关系讨论,最终获得了成药性良好的化合物XY153(8l)。XY153对BRD4 BD2表现出强结合活性,用Octet®检测可以达到nM级别的亲和力,IC50达到0.79 nM,BD2选择性倍数为354倍。该研究为开发针对急性髓性白血病的药物提供了新的候选分子。

抗肿瘤拮抗剂[4]


雄激素受体(AR)是前列腺癌药物研发的重要靶标。靶向AR AF2位点的小分子可以直接阻断共激活因子与AR的结合。该研究通过分子对接联合MM/GBSA重打分双层筛选策略开发了一系列具有N-(4-(苄氧基)-苯基)-磺酰胺骨架的AR AF2拮抗剂。其中,化合物T1-12显示出优异的AR拮抗活性。随后,Octet®小分子蛋白互作分析系统分别测试了小分子T1-12、恩杂鲁胺和SRC2-3多肽分别与AR LBD蛋白和AR LBD-DHT复合物的亲和力,根据体系间亲和力和作用模式差异验证小分子T1-12作用于AR AF2位点。
